Description
Description:
PcrA is an ATP-driven 3′ to 5′ helicase responsible for the unwinding of double-stranded DNA (dsDNA). PcrA binds to and unravels of dsDNA into two strands of single-stranded DNA (ssDNA).
Applications:
Helicase-dependent DNA isothermal amplification
Source:
expressed in E. coli
Organism:
Geobacillus stearothermophilus
Storage:
-20°C
Storage Buffer:
600 mM NaCl, 50 mM Tris pH 7.6, 50% glycerol
10X Reaction Buffer:
500 mM Potassium acetate, 200 mM Tris-acetate, 100 mM Magnesium acetate, 1 mg/ml BSA, pH 7.9
Experiment Result:
Helicase-dependent DNA isothermal amplification reaction (65 °C, 1 hour) using PcrA helicase for cloning 667 bp and 871 bp DNA fragments. Template of the reaction is 5 ng DNA.
